چکیده

A number of different low molecular weight and volatile compounds have been tested and compared as matrices in 252Cf plasma-desorption mass spectrometry (PDMS) measurements of oligosaccharides. Some heteroaromatic amines (Zaminothiazole and 3-aminopyridine), hydroxyanthraquinones (alizarin and quinalizarin) and phthaleins (fluorescein) proved to be especially suited to enhancing the quasimolecular ion intensity of oligosaccharides. Discrimination effects could be reduced by matrix addition and thus, quantitative results of PDMS could be improved. Oligosaccharides with broad molecular weight distribution (dextrin 10 and dextran T 1.5) have been successfully analyzed by PDMS. The mass spectra obtained compared quite well with ion chromatograms of the respective oligosaccharides.

Alkaline degradation of oligosaccharides coupled with matrix-assisted laser desorption/ionization Fourier transform mass spectrometry: a method for sequencing oligosaccharides.

A new technique for determining sequence and linkage information of underivatized oligosaccharides is developed using alkaline degradation and matrix-assisted laser desorption/ionization Fourier transform mass spectrometry (MALDI-FTMS). Alkaline degradation (also known as the "peeling" reaction) is a chemical degradation technique that only cleaves the glycosidic bond at the reducing end by bet...

Electron detachment dissociation of underivatized chloride-adducted oligosaccharides.

Chloride anion attachment has previously been shown to aid determination of saccharide anomeric configuration and generation of linkage information in negative ion post-source decay MALDI tandem mass spectrometry. Here, we employ electron detachment dissociation (EDD) and collision activated dissociation (CAD) for the structural characterization of underivatized oligosaccharides bearing a chlor...

Sequencing of oligosaccharides by collision-induced dissociation matrix-assisted laser desorption/ionization mass spectrometry.

A study of the collision-induced dissociation post-source decay (PSD) spectra of free oligosaccharides is presented. These spectra, when obtained with helium as collision gas, show (1,5)X fragments containing the reducing end sugar. The presence of these fragments permits Y ions and, consequently, B and C peaks to be identified. This is a common behaviour from which it has been possible to deli...

Ionic liquid matrix for direct UV-MALDI-TOF-MS analysis of dermatan sulfate and chondroitin sulfate oligosaccharides.

Polyanionic oligosaccharides such as dermatan sulfate (DS) and chondroitin sulfate (CS) exhibit poor ionization efficiencies and tend to undergo thermal fragmentation through the loss of SO(3) under conventional ultraviolet matrix-assisted laser desorption/ionization (UV-MALDI) conditions. A new ionic liquid matrix (ILM), a guanidinium salt of alpha-cyano-4-hydroxycinnamic acid, facilitates dir...

Californium-252 plasma desorption mass spectrometry of oligosaccharides and glycoconjugates: control of ionization and fragmentation.

The fragmentation of underivatized and peracetylated maltooligosaccharides, ranging in length from four to seven glucose residues, has been investigated by 252Cf plasma desorption mass spectrometry (PDMS). Investigations are made of the effects on the mass spectra of (1) peracetylation, (2) sample preparation by electrospray deposition onto metallic substrates versus adsorption onto nitrocellul...
